EM Monolayer Assay Images
This assay allows us to visualize the early steps of endocytosis
by electron microscopy.
See Ford et al, Science 291: 1051-1055
(.pdf) or Higgins and McMahon,
TIBS 27: 257-259 (.pdf) for details and examples.
Proteins were injected under a 10% PtdIns(4,5)P2 lipid monolayer, incubated at room temperature, stained by uranyl acetate and visualized by EM. (see schematic technique or detailed protocol)
These are images of EM negatives, scale bars of either 100 nm (40000x
images) or 300 nm (20000x images) are visible on the larger images.
The stereo images are best visualized with a separation of 6 cm.
